Asthma PATHOPHYSIOLOGY Asthmatic Bronchial Epithelial Cells Have a Deficient Innate Immune Response to Infection With Rhinovirus

چکیده
METHODS. BECs were cultured from bronchial brushings obtained by bronchoscopy. BECs were studied prerhinovirus-16 infection, postinfection, and post–inactivated rhinovirus-16 exposure. Cytokines and chemokines were measured. Viable cell numbers, numbers of apoptotic cells, and cell lysate caspase activity were used to measure apoptotic activity. Apoptotic activity inhibition using the caspase-3 inhibitor, ZMD-fmk, was used to verify the presence of apoptosis. Rhinovirus-infection induction of interferon(IFN) was measured. Influence of IFNon apoptosis induction was evaluated by prerhinovirus and postrhinovirus infection treatment of BECs with exogenous IFN. The effect of IFNwas also measured on virus titer preinfection and postinfection.

متن کاملAsthmatic bronchial epithelial cells have a deficient innate immune response to infection with rhinovirus
Rhinoviruses are the major trigger of acute asthma exacerbations and asthmatic subjects are more susceptible to these infections. To investigate the underlying mechanisms of this increased susceptibility, we examined virus replication and innate responses to rhinovirus (RV)-16 infection of primary bronchial epithelial cells from asthmatic and healthy control subjects. Viral RNA expression and l...
متن کاملTransforming Growth Factor-Beta Promotes Rhinovirus Replication in Bronchial Epithelial Cells by Suppressing the Innate Immune Response
Rhinovirus (RV) infection is a major cause of asthma exacerbations which may be due to a deficient innate immune response in the bronchial epithelium. We hypothesized that the pleiotropic cytokine, TGF-β, influences interferon (IFN) production by primary bronchial epithelial cells (PBECs) following RV infection. Exogenous TGF-β(2) increased RV replication and decreased IFN protein secretion in ...
متن کاملRhinovirus-induced interferon production in asthma.
After reading the article by Sykes et al, we would like to comment on impaired response to rhinovirus in relation to asthma severity. The authors examined rhinovirus-induced interferon (IFN) production in a cohort of asthmatic patients compared with controls. While they previously reported in this population a reduced IFN production in cells from bronchoalveolar lavage, they fail to document su...
متن کاملDouble-stranded RNA induces disproportionate expression of thymic stromal lymphopoietin versus interferon-b in bronchial epithelial cells from donors with asthma
Background Thymic stromal lymphopoietin (TSLP) is an epithelial cell-derived cytokine that strongly activates dendritic cells and can initiate allergic inflammation. Since exposure to rhinovirus or double-stranded (ds) RNA (a surrogate of viral infection) induces TSLP expression in bronchial epithelial cells (BECs), this cytokine may link innate antiviral responses and the type 2 adaptive immun...
